Properties of DyLight(TM) Fluorescent Dyes.
DyLight(TM) dyes can be used for multi-color immunofluorescence microscopy with uniform fluorescence intensity throughout the image. DyLight(TM) dyes are exceptionally bright and photostable and are optimized for microscopy and microarray detection methods. This image shows anti-histone detection using a DyLight(TM) 488 conjugate (green). Anti-Tubulin was detected using a DyLight(TM) 549 conjugate (red). Nuclei were counter-stained using DAPI (blue). The image was captured using an Axio Imager.Z1 (Zeiss Micro Imaging Inc).
DyLight(TM) 488 Fluorescence Spectra.
Immunohistochemistry. NOS3 increases 3-nitrotyrosine content in heart tissue in developing sepsis. Immunohistochemical staining demonstrated an increase of nitrotyrosine in heart tissue of septic wildtype mice at 6 hours post CLP, whereas no differences could be detected in NOS3-/- mice. Heart samples were fixed in 4% formaldehyde phosphate buffer, dehydrated and paraffin-embedded. For 3-nitrotyrosin detection serial sections (3 per mouse, 400µm apart) were incubated with a polyclonal antibody raised against nitrotyrosine, followed by sheep anti-rabbit IgG conjugated to DyLight(TM)488 (p/n 611-641-122).Figure online resource 1.
