K-975 is a covalent inhibitor of YAP/TAZ-TEAD signaling. It binds to the palmitate-binding pocket of TEAD preventing its association with YAP/TAZ.1 K-975 inhibited proliferation of NF2-non-expressing mesothelioma cell lines. K-975 promoted liver progenitor cell to hepatocyte differentiation via suppression of YAP signaling.2 It sensitized YAP-expressing RET-aberrant cancer cells to RET kinase inhibitors.3
