CRISPR/Cas Genome Editing
The bacterial CRISPR/Cas9 system has been adapted to serve as a versatile platform for RNA-directed genome editing in mammalian cells. The simplicity of using of a noncoding RNA guide to target DNA for site-specific cleavage provides a distinct advantage over alternative genome editing technologies such as ZFNs and TALENs. Using the CRISPR/Cas9 strategy, retargeting the nuclease complex only requires introduction of a new RNA sequence and there is no need to reengineer the specificity of DNA-binding proteins.

RNAi Gene Silencing
The ability to silence genes plays an important role in molecular and cell biology and can be readily applied through transfection. Gene expression can be effectively reduced or eliminated by introducing small noncoding RNA molecules that inhibit RNA translation though a process termed RNA interference (RNAi).
RNA molecules that take part in naturally occurring RNAi pathways include:
- Small interfering RNAs (siRNA) – short (20-25 base pairs) double-stranded RNAs resulting from the cleavage of double-stranded RNAs (dsRNAs)
- MicroRNAs (miRNAs) – a separate class of short single-stranded RNAs (20-22 nucleotides) resulting from the processing of noncoding RNAs

mRNA and Viral RNA Transfection
While smaller RNA species such as siRNA/miRNA, etc. are routinely transfected by researchers investigating gene function, transfection of large RNA molecules such as mRNA transcripts and viral RNA replicons is an emerging trend. mRNA delivery for protein expression offers an inherent advantage as cytoplasmic delivery is sufficient to initiate translation of the transcript.

Co-transfection
Co-transfection or co-delivery of multiple nucleic acids – either similar or different in nature is a technique routinely employed by cell biologists to address increasingly complex experimental design and applications. Common examples of nucleic acid co-transfection are as follows:
- Co-delivery of multiple plasmid DNAs for virus production
- Co-transfection of plasmid DNA and siRNA to investigate gene function
- Co-delivery of plasmid DNA and mRNA for gene insertion, etc.
Co-transfection of different nucleic acid types such as large plasmid DNA and smaller siRNA/miRNAs can be challenging primarily due to the size and charge difference between these nucleic acid molecules. The charge of the nucleic acid molecule impacts the charge ratio of the transfection complexes and their formation and uptake by the mammalian cell membrane. Due to this reason, transfection reagents that can deliver smaller nucleic acids such as siRNA/miRNA to the cytoplasm efficiently are not always successful in plasmid DNA delivery to the nucleus. Unique polymeric formulations such as the TransIT-X2® Dynamic Delivery System are very effective nucleic acid condensing agents.
