Mechanism of Wnt3a stabilization by Afamin:

Wnt3a is a common growth factor used in organoid culture. However, the lipid-soluble form of the Wnt3a protein tends to aggregate in serum-free medium over time, which leads to a loss of activity.

Mihara et al. discovered that Afamin, a serum component, forms a complex with Wnt3a, thereby maintaining its activity. Furthermore, using the Afamin-Wnt3a complex enabled long-term organoid culture.

Intestinal organoid culture using Afamin/Wnt3a CM

Both the new product (art. no. MBL-J2-005-05) and the previous product (art. no. MBL-J2-001) were used to culture human small intestine organoids. At a concentration of 2.5%, the former product was found to decrease organoid growth (see bottom image). Even at an added concentration of 2.5%, the new product maintains robust organoid growth (top).
