Lane 3: Rat IgG Fab Fragment (p/n 012-0105). Lane 4: Rat IgM Whole Molecule (p/n 012-0107). All samples were reduced. Load: 50 ng per lane. Block: MB-070 for 30 min at RT. Primary Antibody: Anti-Rat IgG (H&L) (RABBIT) Antibody (p/n 612-4102) 1:1,000 for 60 min at RT. Secondary Antibody: Anti-Rabbit IgG (GOAT) Peroxidase Conjugated Antibody (p/n 611-103-122) 1:40,000 in MB-070 for 30 min at RT. Predicted/Observed Size: 25 and 55 kDa for Rat IgG, 25 kDa for F(c) and Fab, 78 and 25 kDa for IgM. Rat F(c) migrates slightly higher.
ELISA results of Rabbit Anti-Rat IgG Antibody Peroxidase Conjugated tested against purified Rat IgG protein. Each well was coated in duplicate with 1.0 µg of Rat IgG (p/n 012-0102) [Green Line]. The starting dilution of antibody was 5µg/ml and the X-axis represents the Log10 of a 3-fold dilution. The titer is 1:41,800. This titration is a 4-parameter curve fit where the IC50 is defined as the titer of the antibody. Assay performed using 3% fish gelatin as blocking buffer and TMB substrate p/n TMBE-1000.
