Inducible co-stimulator also called AILIM and CRP-1 (CD28-related protein-1), is a member of the CD28 family of immune co-stimulatory receptors. Other family members are CD28, CTLA-4, and PD-1. Human ICOS is a homodimeric type I transmembrane protein consisting of 199aa with a putative 20aa signal sequence, a 121aa extracellular domain, a 23aa transmembrane region, and a 35aa cytoplasmic domain. ICOS shares ~39% aa similarity with CD28 and CTLA-4. Human and mouse ICOS share ~72% aa identity. ICOS is expressed on most CD45RO+ cells. ICOS expression is up-regulated within ~24-48h of activation on T helper (Th) primed cells. B7-H2, a member of the B7 family of co-stimulatory ligands, has been identified as the ICOS ligand. The B7-H2/ ICOS interaction appears to play roles in T cell-dependent B cell activation and Th differentiation. Applications: Suitable for use in Flow Cytometry. Other applications not tested. Recommended Dilution: Optimal dilutions to be determined by the researcher. Excitation: 620-650nm Emission: 660-670nm Storage and Stability: Store product at 4C in the dark. DO NOT FREEZE! Stable at 4C for 12 months after receipt as an undiluted liquid. Dilute required amount only prior to immediate use. Further dilutions can be made in assay buffer. Caution: APC conjugates are sensitive to light. For maximum recovery of product, centrifuge the original vial prior to removing the cap.
