Tripeptidyl Peptidase I (TPP1), also known as tripeptidyl aminopeptidase, is a lysosomal peptidase which can hydrolyze tripeptides from the Ntermini of oligopeptides and also possesses weak endopeptidase activity (1 3). TPP1 is a serine peptidase with a SerGluAsp catalytic triad, making it a member of the sedolisin family (4). The TPP1 precursor undergoes autoactivation under conditions of acidic pH (4). TPP1 is expressed in many tissues, with elevated expression in tissues associated with peptide hormone production (5). Mutations in TPP1 have been shown to be a cause of classic lateinfantile neuronal ceroid lipofuscinosis (CLN2), a lysosomal storage disease (6). Recombinant corresponding to Met1-Pro563 from human TripeptidylPeptidase I/TPP1, C-terminal, 10-His tag, expressed in NS0 cells. Molecular Weight: ~61kD Biological Activity: Measured by the cleavage of AlaAlaPheAMC. The specific activity is >850 pmoles/min/ug, as measured under the described conditions. Storage and Stability: May be stored at 4C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20C. Aliquots are stable for 6 months after receipt at -20C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
