TFPI-2 (tissue factor pathway inhibitor-2, also placental protein 5/PP5) is a secreted, presumably 27-34kD glycoprotein member of the Kunitz domain-containing proteinase inhibitor family. In mouse, it is principally produced by endothelium, hepatocytes and synctiotrophoblasts, and serves as an inhibitor of multiple serine proteinases. Mature mouse TFPI-2 is 208aa in length. It contains three Kunitz inhibitor domains, the most N-terminal of which (aa36-86) is crucial to inhibitor activity. Over aa23-211, mouse TFPI-2 shares 86% and 58% aa sequence identity with rat and human TFPI-2, respectively. Applications: Suitable for use in ELISA, Immunohistochemistry and Western Blot. Other applications not tested. Recommended Dilution: Immunohistochemistry (Formalin fixed paraffin embedded): 1ug/ml Optimal dilutions to be determined by the researcher. For long-term storage, aliquot to avoid repeated freezing and thawing and store at -20C. Aliquots are stable at -20C for 12 months after receipt. Dilute required amount only prior to immediate use. Further dilutions can be made in assay buffer. Note: Sodium azide is a potent inhibitor of peroxidase and should not be added to HRP conjugates. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Note: Applications are based on unconjugated antibody.
