Murine antibodies were produced by immunising BALB/c mice with the mouse erythroleukaemic (MEL) cell line stably expressing high levels of wildtype, full-length human Kell protein. This overexpression allowed several epitopes of Kell protein to serve as the immunogen, resulting in identification of several anti-Kell mAbs with different specificities, of which MIMA-8 targeted the Jsb antigen. Splenocytes were obtained from immunised mice and fused with the P3X63Ag8.653 myeloma cell line to produce a hybridoma.
