ICAD/DFF45 is an inhibitor of the caspase-activated DNase (CAD, DFF40). DNA fragmentation factor (DFF) consists of two subunits, a 40 kDa caspase-activated nuclease (DFF40) and a 45 kDa inhibitor (DFF45). In proliferating cells CAD or DFF40 forms a complex with ICAD or DFF45. When cells undergo apoptosis, caspase 3 cleaves ICAD at two sites to dissociate the CAD:ICAD complex, allowing CAD to cleave chromosomal DNA. Thus DEF induces the fragmentation of DNA which is a hallmark of apoptosis. ICAD is expressed in brain, blood, skin, spleen, adipose tissue. It has 2 isoforms produced by alternative splicing.
