In the assay, esterase catalyzes the hydrolysis of cholesterol esters to produce free cholesterol (FC) and free fatty acids (FFA). Then Cholesterol oxidase catalyzes free cholesterol (FC) to produce 4-cholestenone and H2O2. Further, H2O2, 4-aminoantipyrine and phenol can be catalyzed by peroxidase to form red quinone compounds which has a characteristic absorption peak at 500 nm.
