Application Notes: Caspase-12 (large) antibody can be used for the detection of both the propeptide and cleaved forms of caspase-12 by Western blot at 0.25 to 2 µg/mL. Antibody can also be used for immunohistochemistry starting at 2 µg/mL. For immunofluorescence start at 10 µg/mL.Antibody validated: Western Blot in human, mouse, and rat samples, Immunohistochemistry in human, mouse, and rat samples and Immunofluorescence in human, mouse, and rat samples. All other applications and species not yet tested

Immunofluorescence Validation of Caspase-12 in Mouse Heart. Immunofluorescent analysis of 4% paraformaldehyde-fixed mouse heart labeling caspase-12 with orb1239263 at 20 µg/mL, followed by goat anti-rabbit IgG secondary antibody at 1/500 dilution (green) and DAPI antibody (blue).

Immunohistochemistry Validation of Caspase-12 in Human Colon. Immunohistochemical analysis of paraffin-embedded human colon tissue using anti-Caspase-12 antibody (orb1239263) at 1 µg/mL. Tissue was fixed with formaldehyde and blocked with 10% serum for 1 h at RT, antigen retrieval was by heat mediation with a citrate buffer (pH6). Samples were incubated with primary antibody overnight at 4C. A goat anti-rabbit IgG H&L (HRP) at 1/250 was used as secondary. Counter stained with Hematoxylin.
