AH109 is a MATa type strain used in GAL4 system yeast hybrid experiments, which can be directly transformed into plasmids or for protein interaction validation or library screening with MATa type yeast strain Y187 through mating. The Transformation markers are: trpl, leu2, and reporting genes are: lacZ, HIS3, ADE2, MEL1. The AH109-GAL4 yeast hybrid system requires two plasmids to be used in combination: pGBKT7 and pGADT7. The screening marker for plasmid pGBKT7 is TRP1, used to express the fusion protein of DNA-BD (amino acids 1-174 from the GAL4N end of yeast transcription factor) and target protein (Bait). The screening marker for plasmid pGADT7 is LEU, used to express the fusion protein of AD (amino acids 768-881 from the GAL4C end) and target protein (Prey).
