Herpes simplex virus protein 16 (VP16) is a transcription activator that can activate downstream gene expression. The transcriptional activation region of VP16 was fused and expressed with GAL4-BD to construct the pGBKT7-VP16 (BD-VP16) vector, which was then transformed into Y2HGold. The transcriptional activation of VP16 will activate the expression of reporter genes HIS3 and ADE2 in yeast, enabling normal growth on histidine and adenine deficient media. Transcription factor X is fused and expressed with BD and BD-VP16, respectively. If transcription factor X activates downstream genes, it will activate the expression of downstream genes. Compared with the control strain containing BD, the strain containing BD-X can grow on defective culture medium. If transcription factor X exerts transcriptional repression on downstream genes, it will partially inhibit the activation of downstream genes by VP16. Compared with the control strain containing BD-VP16, the strain containing BD-VP16-X inhibits the activation of downstream reporter genes, thereby inhibiting growth on defective culture medium.
