Many site-specific transcriptional activation factors in eukaryotes typically have two separable domains, namely the DNA binding domain (BD) and the transcriptional activation domain (AD). These two domains have their own functions and do not affect each other. But a complete activation factor that activates specific gene expression must contain both domains, otherwise it cannot complete the activation function. Based on this principle, two test proteins can be fused with these two domains to form fusion proteins and co expressed in the same yeast cell. If two test proteins can interact with each other, the bridging effect of the test proteins will enable AD and BD to form a complete transcriptional activation factor and activate the corresponding reporter gene expression. It is easy to determine whether there is any interaction between the molecules of the tested protein system by measuring the phenotype of the reported gene.
