The Y1H pAbAi system is a commonly used for validation the interaction between promoter functional elements and transcription factors, or screening upstream regulatory proteins of promoter functional elements. The vectors used for the Y1H pAbai system are pAbAi and pGADT7, and the host is Y1HGold. The system usually selects the promoter acting element, designs and replicates it three times to construct into the pAbAi vector, with a length generally below 200bp. The advantage is that the interacting genes can be proven to directly interact with the elements, but because only one element is selected at a time, relatively fewer proteins are screened out or the probability of verifying the interaction is lower. This kit contains the basic culture medium and reagents required for library screening by Y1H pAbAi system.
