EGY48 yeast can be used for both single and two hybrid experiments, plasmid transformation for interaction verification or library screening experiments, with screening markers of his3, trp1, ura3. The EGY48-LacZ yeast hybrid system requires the use of two plasmids, pB42AD and pLacZi-2u. The screening marker for plasmid pB42AD is TRP1, which is used to express the fusion protein of AD (B42AD protein composed of 88 amino acid residues from herpes virus) and the target protein (Prey). The screening marker for plasmid pLacZi is URA3, and the reporter gene is LacZ. LacZ expression can only be initiated when Bait and Prey interact, and it turns blue on chromogenic medium, thus confirming the interaction between DNA and protein. This product combines the LacZ gene extracellular expression colorimetric kit for interaction validation. The dilution point plate method is used, which can more intuitively reflect the interaction between DNA and protein compared to the plate coating method, and can also reduce the use of culture medium. In addition, this kit also also validates the interaction of positive clones.
