Tumor protein p53, also known as p53, cellular tumor antigen p53 (UniProt name), phosphoprotein p53, tumor suppressor p53, antigen NY-CO-13, or transformation- related protein 53 (TRP53), is any isoform of a protein encoded by homologous genes in various organisms, such as TP53 (humans) and Trp53 (mice). TP53 involved in cell cycle regulation as a trans-activator that acts to negatively regµl ate cell division by controlling a set of genes required for this process. One of the activated genes is an inhibitor of cyclin-dependent kinases. To test the effect of TP53 on cell apoptosis, Jurkat cells were seeded into triplicate wells of 96-well plates at a density of 5000 cells/well with 1% serum standard RPMI-1640 including various concentrations of recombinant human TP53. After incubated for 72h, cells were observed by inverted microscope and cell proliferation was measured by Cell Counting Kit-8 (CCK-8). Briefly, 10 µl of CCK-8 solution was added to each well of the plate, then the absorbance at 450nm was measured using a microplate reader after incubating the plate for 1-4 hours at 37C. Proliferation of Jurkat cells after incubation with TP53 for 72h observed by inverted microscope.
