MDANP (Milk-derived anti-NEC peptide) is a peptide identified from human milk. Bioactive peptides are an important component of human milk and they exhibit a variety of functions such as anti-infection, immune regulation, and growth promotion. Endoplasmic reticulum (ER) stress plays a critical role in necrotizing enterocolitis (NEC) pathogenesis through the PERK-eIF2E'-QRICH1 axis, and in an effort to identify agents in milk which can ameliorate it, MDANP is able to improve intestinal cell apoptosis and has a protective effect in mouse models. That reduction of ER stress by MDANP acts through the PERK-eIF2 alpha-QRICH1 pathway is shown by MDANP reducing the levels of PERK, p-eIF2, and QRICH1 proteins. MDANP both decreases the severity of NEC in neonatal mouse and ameliorates intestinal injury and reduces apoptosis.
