AK59 is a STING molecular glue degrader that functions by leveraging HERC4, a hect domain E3 ligase. AK59 forms a ternary complex between STING and HERC4, leading to the ubiquitination of STING at Lys150 and subsequent degradation by the proteasome. AK59S effectively inhibits downstream signal transduction, including the phosphorylation of TBK1 and IRF3, as well as the production of cytokines such as IFN-beta and CXCL10. AK59 is applicable to the research of autoimmune diseases[1].
