ISM5939 is an orally active ENPP1 inhibitor. By potently and selectively inhibiting ENPP1, ISM5939 blocks its degradation of extracellular cGAMP, thereby stabilizing and enriching cGAMP in the tumor microenvironment. ISM5939 locally and continuously activates the STING pathway, induces type I interferon production, remodels the immunosuppressive tumor microenvironment, enhances anti-tumor immune responses, while avoiding side effects such as systemic inflammation and T-cell death caused by direct STING agonists. ISM5939 can be used in the research of solid tumors[1].
