ATTO 647 is a carborhodamine fluorophore and imaging/tracking agent.[1][2]ATTO 647 binds to N-acetyl-beta-D-glucosaminyl and sialic acid residues on membrane glycoproteins when conjugated to wheat germ agglutinin, enabling single-molecule tracking of glycoprotein diffusion.[1][2]ATTO 647 serves as a fluorescent probe for single-molecule fluorescence tracking studies of cell membrane structure and diffusional properties.[1][2]ATTO 647 acts as a fluorescence stabilizer with reduced blinking and stable fluorescence in environments with sufficient oxidant and reductant concentrations, and variable blinking and fluorescence profiles across different mounting media.[1][2]ATTO 647 acts as a photostability enhancer with enhanced photostability in Ibidi-MM, ROXS (AA/MV), and ROXS (TX/TQ) mounting media.[1][2]ATTO 647 labels histone H2B-GFP in fixed cells for confocal microscopy photobleaching experiments.[1][2]ATTO 647 shows preserved brightness in fresh ROXS (AA/MV) and ROXS (TX/TQ) samples, with moderate brightness decrease after 3 days at 4 C, and variable brightness profiles in Vectashield and Ibidi-MM samples.[1][2]ATTO 647 exhibits extremely stable fluorescence in ROXS (AA/MV) and ROXS (TX/TQ) buffers during single-molecule experiments with immobilized DNA.[1][2]
