Z-DEVD-AFC is a specific fluorescent substrate widely used in apoptosis (Apoptosis) research, primarily for the quantitative detection of the protease activity of Caspase-3 (as well as Caspase-7, which has similar substrate specificity). Z-DEVD-AFC itself shows weak fluorescence (or exists in a quenched/low quantum yield state). When apoptosis occurs in cells, activated Caspase-3/7 precisely cleaves the peptide bond of the Asp residue at the C-terminus of the DEVD sequence, releasing free AFC groups (characteristics of free AFC: Ex/Em ~ 400 nm/505 nm)[1][2].
