The HERC2 protein is an E3 ubiquitin protein ligase that tightly regulates ubiquitin-dependent retention of repair proteins on damaged chromosomes, especially in response to ionizing radiation (IR). HERC2 is recruited to DNA damage sites and promotes the assembly of UBE2N and RNF8, promoting Lys-63 linked ubiquitin chains to produce DNA damage-induced responses.
