Interaction with MAPKAPK5 leads to ERK3/MAPK6 phosphorylation at Ser-189, activating MAPKAPK5, which reciprocally phosphorylates ERK3/MAPK6. The specific cellular role of this interplay, while unclear, suggests ERK3s potential involvement in promoting entry into the cell cycle.
