SMPDL3A protein hydrolyzes nucleotide triphosphates, particularly ATP, and p-nitrophenyl-TMP. It has limited activity with nucleoside diphosphates and none with nucleoside monophosphates. SMPDL3A also catalyzes the conversion of CDP-choline to CMP and phosphocholine, and CDP-ethanolamine. However, it lacks sphingomyelin phosphodiesterase activity.
