DA-143 is a selective inhibitor of DNA-PKcs (IC50: 2.5 nM), demonstrating disparate inhibitory effects on mTOR, PI3Kdelta, and ATM, with IC50 values of 280 nM, 106 nM, and 6,594 nM, respectively. This compound effectively blocks the phosphorylation of DNA-PKcs substrates and enhances the sensitivity of cancer cells to doxorubicin.
