Sofnobrutinib (AS-0871) is an orally active, selective inhibitor of Brutons tyrosine kinase (BTK) with IC50 values of 4.2 nM for activated BTK and 0.39 nM for unactivated BTK. It has an IC50 of 24 µM against hERG, exhibits strong metabolic stability and bioavailability, induces a weak passive cutaneous anaphylaxis reaction, and demonstrates anti-inflammatory properties in a mouse model of collagen-induced arthritis.
