Detection Range: 246.9-20,000pg/ml Sensitivity: 95.6pg/ml Intra-Assay: CV<10% Inter-Assay: CV<12% Kit Components: *025275A: Microtiter Strips, 1x96 wells *025275B: Standard, 2x1vial 025275C: Standard Diluent, 1x20ml *025275D: Detection Reagent A (green), 1x120ul *025275E: Detection Reagent B (red), 1x120ul 025275F: Assay Diluent A, 1x12ml 025275G: Assay Diluent B, 1x12ml 025275H: TMB Substrate, 1x9ml *025275J: Positive Control, 1094.2ng/ml, 1x1vial 025275K: Stop Solution, 1x6ml 025275L: Wash Buffer, 30x, 1x20ml Precaution: The Stop Solution (025275K) suggested for use with this kit is an acid solution. Wear eye, hand, face, and clothing protection when using this material. Storage and Stability: Store *025275A, *025275B, *025275D, *025275E and *025275J at -20C. Store all the other components at 4C. After reconstitution of *025275J, store at 4C and use within 5 days. Store all the other components at 4C. Unused kit is stable for 6 months after receipt. Once kit components are opened, it is highly recommended to use remaining reagents within 1 month provided this is within the expiration date of the kit. For maximum recovery of product, centrifuge the original vials after thawing and prior to removing the cap. Notes on 025275J: Positive Control: Intended Use: The control vial is from plant GA. It will provide a positive result in the ELISA assay, which indicates that the kit is optimized and working properly. Characteristics: The control vial containsplant GA from plant leaves tissues (lyophilized powder). Expected Values: 1094.2ng/ml Usage: Dissolve with 150ul standard diluent and let it stand for 10 minutes. If powder is partly insoluble, shake gently until fully dissolved. Mix gently before use. In the experiment, add 100ul to an appropriate well of the plate and test its concentration along with standards and your specimens. Note: Upon receipt store Positive Control vial at -20C. After reconstitution, store Positive Control vial at 4C and use within 5 days. Assay Procedure Summary: 1. Prepare all reagents, samples and standards. 2. Add 50ul standard or sample to each well, then add 50ul prepared Detection Reagent A immediately. Shake and mix. Incubate 1 hour at 37C. 3. Aspirate and wash 3 times. 4. Add 100ul prepared Detection Reagent B. Incubate 30 minutes at 37C. 5. Aspirate and wash 5 times. 6. Add 90ul Substrate Solution. Incubate 15-25 minutes at 37C. 7. Add 50ul Stop Solution. Read at 450nm immediately.
