Cysteinerich with EGFlike domain protein 1 (CRELD1) is an ~50kD transmembrane glycoprotein that contains a 333aa extracellular domain (ECD), two tandem transmembrane segments, and a second ECD of 15aa (1). Within the large ECD, mouse and human CRELD1 share 92%aa sequence identity. CRELD1 is widely expressed during development and in the adult (1). It interacts with Reticulon 3/RTN3 and inhibits the proapoptotic activity of RTN3 (2). CRELD1 also associates with Adrenergic alpha1 receptors in the prostate and limits their expression (3). Missense mutations of human CRELD1 are associated with susceptibility to atrioventricular septal defects in the heart (4). Molecular Weight: ~37.1kD Biological Activity: Measured by its ability to induce adhesion of ATDC5 mouse chondrogenic cells. The ED50 for this effect is typically 0.3-1.5ug/ml. Endotoxin: <0.10 EU per 1ug of protein by the LAL method Storage and Stability: Lyophilized powder may be stored at -20C. Stable for 12 months at -20C. Reconstitute with sterile buffer. Aliquot to avoid repeated freezing and thawing. Store at -20C. Reconstituted product is stable for 6 months at -20C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
