The Microalbuminuria (MAU/ALB), BioAssay(TM) ELISA Kit is a sandwich ELISA intended for the quantitative determination of rat Microalbuminuria (MAU/ALB) concentrations in serum, plasma and urine. Detection Range: 0.01-40ug/ml Sensitivity: 0.01ug/ml Specificity: This assay has high sensitivity and excellent specificity for detection of rat MAU/ALB. No significant cross-reactivity or interference between rat MAU/ALB and analogues was observed. Precision: Intra-assay: CV% <8% Inter-assay: CV% <10% Principle of the Assay: This BioAssay(TM) employs the quantitative sandwich enzyme immunoassay technique. An antibody specific for MAU/ALB has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any MAU/ALB present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for MAU/ALB is added to the wells. After washing, avidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of MAU/ALB bound in the initial step. The color development is stopped and the intensity of the color is measured. Kit Components: 227812A: Microtiter Strips, 1x48 wells or 1x96 wells. Pre-coated, ready to use. 227812B: Standard, 100X, 1x100ul 227812C: MAU/ALB (Biotin), 100X, 1x60ul or 1x120ul 227812D: Avidin (HRP), 100X, 1x60ul or 1x120ul 227812E: Biotin Diluent, 1x5ml or 1x10ml 227812F: Avidin (HRP) Diluent, 1x10ml or 1x20ml 227812G: Sample Diluent, 2x20ml 227812H: Wash Buffer, 25X, 1x10ml or 1x20ml 227812J: TMB Substrate, 1x5ml or 1x10ml 227812K: Stop Solution, 1x5ml or 1x10ml Note: The Stop Solution (227812K) provided with this kit is an acid solution. Wear eye, hand, face, and clothing protection when using this material. Storage and Stability: Store kit at 4C. The unopended kit is stable for 6 months after receipt. Once opened, the kit is stable up to 4 weeks at 4C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Assay Summary: 1. Prepare reagents, standards and samples as instructed. 2. Add 100ul standard or sample to each well. Incubate for 1 hour at 37C 3. Aspirate and wash 3 times. 4. Add 100ul MAU/ALB (Biotin), 1X to each well. Incubate for 1 hour at 37C 5. Aspirate and wash 3 times. 6. Add 100ul Avidin (HRP), 1X to each well. Incubate for 1 hour at 37C 7. Aspirate and wash 5 times. 8. Add 90ul TMB substrate to each well Incubate for 20 minutes at 37C. Protect from light 9. Add 50ul Stop Solution to each well. Read at 450nm within 5 minutes.
