Thyroxine (T4), BioAssay(TM) ELISA Kit is a competitive ELISA intended for the quantitative determination of Thyroxine (T4) in fish serum, plasma and tissue homogenates. Detection Range: 20-320ng/ml Intra-assay Precision: CV <15% Inter-assay Precision: CV <15% Sensitivity: The minimum detectable dose of fish Thyroxine (T4) is typically less than 20ng/ml. The sensitivity of this assay, or Lower Limit of Detection (LLD) was defined as the lowest fish Thyroxine (T4) concentration that could be differentiated from zero. It was determined the mean O.D value of 20 replicates of the zero standard added by their three standard deviations. Specificity: This assay has high sensitivity and excellent specificity for detection of fish Thyroxine (T4). No significant crossreactivity or interference between fish Thyroxine (T4) and analogs was observed. Note: Limited by current skills and knowledge, it is impossible for us to complete the crossreactivity detection between fish Thyroxine (T4) and all the analogs. Therefore, crossreaction may still exist. Kit Components: 227960A: Microtiter Strips, 1x48 wells or 1x96 wells 227960B: Standard, 20ng/ml, 1x500ul or1x1ml 227960C: Standard, 40ng/ml, 1x500ul or 1x1ml 227960D: Standard, 80ng/ml, 1x500ul or 1x1ml 227960E: Standard, 160ng/ml, 1x500ul or 1x1ml 227960F: Standard, 320ng/ml, 1x500ul or 1x1ml 227960G: T4 (Biotin), 1x3ml or 1x6ml 227960H: Avidin (HRP) 1x3ml or 1x6ml 227960J: Wash Buffer, 20X, 1x15ml 227960K: Substrate A, 1x3.5ml or 1x7ml 227960L: Substrate B, 1x3.5ml or 1x7ml 227960M: Stop Solution, 1x3.5ml or 1x7ml Note: The Stop Solution (227960M) provided with this kit is an acid solution. Wear eye, hand, face, and clothing protection when using this material. Storage and Stability: Store all kit components at 4C. Stable for 6 months after receipt. Once opened, the kit is stable for 1 month providing this is within the expiration date of the kit. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Assay Summary: 1. Prepare reagents and samples as instructed. 2. Set a Blank well without any solution 3. Add 50ul Standard or sample to each well. 4. Add 50ul T4 (Biotin) to each well (Not to the Blank well). 5. Incubate for 60 minutes at 37C. 6. Aspirate and wash 3 times. 7. Add 50ul Avidin (HRP) to each well (Not to the Blank well). 8. Incubate for 30 minutes at 37C. 9. Aspirate and wash 3 times, 10. Add 50ul Substrate A and 50ul Substrate B to each well. 11. Incubate for 15 minutes at 37C. Protect from light 12. Add 50ul Stop Solution to each well. Read at 450nm within 10 minutes.
