Tri-Iodothyronine (T3), BioAssay(TM) ELISA Kit is a competitive ELISA intended for the quantitative determination of Tri-Iodothyronine in porcine serum and plasma. Detection Range: 500-8000pg/ml Intra-assay Precision: CV <15% Inter-assay Precision: CV <15% Sensitivity: The minimum detectable dose of porcine T3 is typically less than 500pg/ml. The sensitivity of this assay, or Lower Limit of Detection (LLD) was defined as the lowest porcine T3 concentration that could be differentiated from zero. It was determined the mean O.D value of 20 replicates of the zero standard added by their three standard deviations. Specificity: This assay has high sensitivity and excellent specificity for detection of porcine T3. No significant crossreactivity or interference between porcine T3 and analogs was observed. Note: Limited by current skills and knowledge, it is impossible for us to complete the crossreactivity detection between porcine T3 and all the analogs. Therefore, crossreaction may still exist. Wear eye, hand, face, and clothing protection when using this material. Storage and Stability: Store all kit components at 4C. Stable for 6 months after receipt. Once opened, the kit is stable for 1 month providing this is within the expiration date of the kit. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Assay Summary: 1. Prepare reagents and samples as instructed. 2. Set a Blank well without any solution 3. Add 50ul Standard or sample to each well. 4. Add 50ul T3 (Biotin) to each well (Not to the Blank well). 5. Incubate for 60 minutes at 37C. 6. Aspirate and wash 3 times. 7. Add 50ul Avidin (HRP) to each well (Not to the Blank well). 8. Incubate for 30 minutes at 37C. 9. Aspirate and wash 3 times, 10. Add 50ul Substrate A and 50ul Substrate B to each well. 11. Incubate for 15 minutes at 37C. Protect from light 12. Add 50ul Stop Solution to each well. Read at 450nm within 10 minutes.
