Pustulan is a beta-1,6-glucan isolated from Lasallia pustulata MW (20kD). It is recognized by the membrane bound Dectin-1, a C-type lectin-like pattern recognition receptor. Detection of beta-glucans by Dectin-1 receptor leads to the CARD9-dependent activation of NF-kappaB and MAP kinases. Studies have shown that pustulan can stimulate innate immune responses, inducing heat shock protein expression, eliciting phagocytosis, and production of pro-inflammatory cytokines. Source: Lasallia pustulata Specificity: Dectin-1 ligand Synonyms: beta(1-6) linked D-glucose polysaccharide CAS No: 37331-28-5 Molecular Formula: 20kD Molecular Weight: 20kD Purity: 80% Appearance: White to light gray to brown powder 1H-NMR: Conforms to reference Preparation of Pustulan Suspension (1mg/ml): Stimulation of Dectin-1 can be achieved with 1-100mg/ml of pustulan. 1. Weigh 10mg of pustulan in a round-bottom tube. 2. Add 10ml of water to the 10mg of pustulan. Vortex to homogenize. 3. Dispense water in a single expulsion to avoid the formation of clumps. Note: Pustulan is insoluble and results in a non-homogeneous suspension with gelatinous precipitates. Avoid the use of conical tubes. Storage and Stability: Store at 4C under inert atmosphere. For maximum recovery of product, centrifuge the original vial prior to removing the cap.
