Glk belongs to the bacterial glucokinase family. This protein is not highly important in E. coli as glucose is transported into the cell by the PTS system already as glucose 6-phosphate. Source: Recombinant protein corresponding to aa1-321 from E. coli glk, fused to His-tag at N-terminal expressed in E. coli. Molecular Weight: ~37.1kD (344aa) Specific Activity: >70units/mg, obtained by measuring the increase of NADPH in absorbance at 340nm resulting from the reduction of NADP. One unit will oxidize 1umole of Glucose to D-glucose 6-phosphate/minute in the presence of Beta-NADP at pH 9.0 at 37C. Aliquot to avoid repeated freezing and thawing.. Store at -20C. Aliquots are stable for 6 months after receipt at -20C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
