Alpha-2-macroglobulin is a protease inhibitor and cytokine transporter. It inhibits many proteases, including trypsin, thrombin and collagenase. A2M is implicated in Alzheimer disease (AD) due to its ability to mediate the clearance and degradation of A-beta, the major component of beta-amyloid deposits. Sample Type: Serum, plasma, tissue homogenates Intended Use: For the quantitative determination of human a2 macroglobulin (a2 MG) concentrations in serum, plasma and tissue homogenates. Sensitivity: 1.25ug/ml Range: 1.25-20ug/ml Specificity: This assay has high sensitivity and excellent specificity for detection of human a2 MG. No significant cross-reactivity or interference between human a2 MG and analogues was observed. Test Principle: This BioAssay(TM) employs the competitive inhibition enzyme immunoassay technique. The microtiter plate provided in this kit has been pre-coated with a2 MG. Standards or samples are added to the appropriate microtiter plate wells with Horseradish Peroxidase (HRP) conjugated antibody preparation specific for a2 MG. The competitive inhibition reaction is launched between with pre-coated a2 MG and a2 MG in samples. A substrate solution is added to the wells and the color develops in opposite to the amount of a2 MG in the samples. The color development is stopped and the intensity of the color is measured.
