This protein is a type I transmembrane glycoprotein which shows homology to the pMEL17 precursor, a melanocyte-specific protein. GPNMB shows expression in the lowly metastatic human melanoma cell lines and xenografts but does not show expression in the highly metastatic cell lines. GPNMB may be involved in growth delay and reduction of metastatic potential. Two transcript variants encoding different isoforms have been found for this gene. No significant cross-reactivity or interference between human GPNMB and analogues was observed. Test Principle: This BioAssay(TM) employs the quantitative sandwich enzyme immunoassay technique. Antibody specific for GPNMB has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any GPNMB present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for GPNMB is added to the wells. After washing, avidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of GPNMB bound in the initial step. The color development is stopped and the intensity of the color is measured.
