This protein is a member of the TNF-receptor superfamily. This receptor has been found to be essential in mediating a broad variety of immune and inflammatory responses including T cell-dependent immunoglobulin class switching, memory B cell development, and germinal center formation. AT-hook transcription factor AKNA is reported to coordinately regulate the expression of this receptor and its ligand, which may be important for homotypic cell interactions. Adaptor protein TNFR2 interacts with this receptor and serves as a mediator of the signal transduction. The interaction of this receptor and its ligand is found to be necessary for amyloid-beta-induced microglial activation, and thus is thought to be an early event in Alzheimer disease pathogenesis. No significant cross-reactivity or interference between mouse CD40 and analogues was observed. Test Principle: This BioAssay(TM) employs the quantitative sandwich enzyme immunoassay technique. Antibody specific for CD40 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any CD40 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for CD40 is added to the wells. After washing, avidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of CD40 bound in the initial step. The color development is stopped and the intensity of the color is measured.
