This protein is a member of the TNF-receptor superfamily. This protein is one of the major receptors for the tumor necrosis factor-alpha. This receptor can activate NF-kappaB, mediate apoptosis, and function as a regulator of inflammation. Antiapoptotic protein BCL2-associated athanogene 4 (BAG4/SODD) and adaptor proteins TRADD and TRAF2 have been shown to interact with this receptor, and thus play regulatory roles in the signal transduction mediated by the receptor. Germline mutations of the extracellular domains of this receptor were found to be associated with the autosomal dominant periodic fever syndrome. The impaired receptor clearance is thought to be a mechanism of the disease. Sample Type: Serum, plasma, tissue homogenates Intended Use: For the quantitative determination of mouse tumor necrosis factor soluble receptor I (TNFsR-I) concentrations in serum, plasma and tissue homogenates. Sensitivity: 1.261pg/ml Range: 15.625-1000pg/ml Specificity: This assay has high sensitivity and excellent specificity for detection of mouse TNFsR- I . No significant cross-reactivity or interference between mouse TNFsR-I and analogues was observed. Test Principle: This BioAssay(TM) employs the quantitative sandwich enzyme immunoassay technique. Antibody specific for TNFsR-I has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any TNFsR-I present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for TNFsR-I is added to the wells. After washing, avidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of TNFsR-I bound in the initial step. The color development is stopped and the intensity of the color is measured.
