Atrial natriuretic peptide-converting enzyme, also called CRN or ATC2, is a protein that in humans is encoded by the CORIN gene. It is mapped to 4p12. This gene encodes a member of the type II transmembrane serine protease class of the trypsin superfamily. Members of this family are composed of multiple structurally distinct domains. It has been found that Corin is the pro-ANP-converting enzyme and that Corin-mediated pro-ANP activation may play a role in regulating blood pressure. Corin and ANP are essential for physiologic changes at the maternal-fetal interface. This protein may also function as a pro-brain-type natriuretic peptide convertase. Corin gene mutations which cause Corin to impair activity might be an underlying mechanism in hypertension. A monoclonal antibody from mouse specific for Corin has been precoated onto 96-well plates. Standards(NSO, S67-N1042) and test samples are added to the wells, a biotinylated detection polyclonal antibody from goat specific for Corin is added subsequently and then followed by washing with PBS or TBS buffer. Avidin-Biotin-Peroxidase Complex was added and unbound conjugates were washed away with PBS or TBS buffer. HRP substrate TMB was used to visualize HRP enzymatic reaction. TMB was catalyzed by HRP to produce a blue color product that changed into yellow after adding acidic stop solution. The density of yellow is proportional to the human Corin amount of sample captured in plate.
