FGF19, Fibroblast growth factor 19, is a protein that in humans is encoded by the FGF19 gene. The protein encoded by this gene is a member of the fibroblast growth factor(FGF) family. The FGF19 gene is mapped to 11q13.3. The deduced 216aa FGF19 protein contains asignal sequence and 2 cysteine residues that are conserved in the FGF family. Expression of this gene was detected only in fetal but not adult brain tissue. Synergistic interaction of the chick homolog and Wnt-8c has been shown to be required for initiation of inner ear development. FGF19 stimulates hepatic protein and glycogen synthesis but does not induce lipogenesis. The effects of FGF19 are independent of the activity of either insulin or the protein kinase Akt and, instead, are mediated through a mitogen-activated protein kinase signaling pathway that activates components of the protein translation machinery and stimulates glycogen synthase activity. Sample Type: Cell culture supernates, serum and plasma (heparin, EDTA) Intended Use: For quantitative detection of human FGF19 in cell culture supernates, serum and plasma (heparin, EDTA). Sensitivity: <10pg/ml Range: 15.6-1000pg/ml Specificity: Natural and recombinant human FGF19 Test Principle: This human FGF19 BioAssay(TM) ELISA Kit was based on standard sandwich enzyme-linked immune-sorbent assay technology. A monoclonal antibody from mouse specific for FGF19 has been precoated onto 96-well plates. Standards(E.coli) and test samples are added to the wells, a biotinylated detection polyclonal antibody from goat specific for FGF19 is added subsequently and then followed by washing with PBS or TBS buffer. Avidin-Biotin-Peroxidase Complex was added and unbound conjugates were washed away with PBS or TBS buffer. HRP substrate TMB was used to visualize HRP enzymatic reaction. TMB was catalyzed by HRP to produce a blue color product that changed into yellow after adding acidic stop solution. The density of yellow is proportional to the human FGF19 amount of sample captured in plate.
