Interleukin 17D (IL-17D) is a member of the unique IL-17 family, which share a conserved cysteine motif. It is mapped to 13q12.11. IL-17D regulates cytokine production in endothelial cells and inhibits hemopoiesis in vitro. Purified recombinant IL-17D inhibited myeloid progenitor cell colony formation. IL-17D can enhance endothelial cell production of IL6, IL8, and GMCSF, but not other cytokines. And it is preferentially expressed in skeletal muscle, brain, adipose tissue, heart, lung, and pancreas. Sample Type: Cell culture supernates, serum and plasma (heparin, EDTA) Intended Use: For quantitative detection of mouse IL-17D in cell culture supernates, serum and plasma (heparin, EDTA). Sensitivity: <10pg/ml Range: 62.5-4000pg/ml Specificity: Natural and recombinant mouse IL-17D Test Principle: This mouse IL-17D BioAssay(TM) ELISA Kit was based on standard sandwich enzyme-linked immune-sorbent assay technology. A monoclonal antibody from rat specific for IL-17D has been precoated onto 96-well plates. Standards(E.coli) and test samples are added to the wells, a biotinylated detection polyclonal antibody from goat specific for IL-17D is added subsequently and then followed by washing with PBS or TBS buffer. Avidin-Biotin-Peroxidase Complex was added and unbound conjugates were washed away with PBS or TBS buffer. HRP substrate TMB was used to visualize HRP enzymatic reaction. TMB was catalyzed by HRP to produce a blue color product that changed into yellow after adding acidic stop solution. The density of yellow is proportional to the mouse IL-17D amount of sample captured in plate.
