Source: Recombinant protein corresponding to aa1-486 from human Kynurenine 3-Monooxygenase, fused to His-FLAG-Tag at C-terminal, expressed in Baculovirus infected Sf9 cell. Molecular Weight: ~58kD Specific Activity: ~100pmol/min/ug Assay Conditions: KMO was tested with various amounts of enzyme. The reactions in KMO Assay Buffer (50mM sodium phosphate, pH 7.5, 0.1% (w/v) Brij35) contains 200uM NADPH, 400uM LKynurenine, and various concentrations of KMO. The reaction mixture incubated for 90 minutes while reading the UV absorption signal. For the negative control (blank), assay buffer was added instead of the enzyme. Absorption signals at 340nm were measured using a Tecan Infinite M1000 plate reader. Applications: Suitable for use in the study of enzyme kinetics, screening inhibitors, and selectivity profiling. Storage and Stability: May be stored at 4C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20C. Aliquots are stable for 6 months after receipt at -20C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
