Heparinase III prevents the formation of new blood vessels in vivo.[1] It specifically degrades heparin sulphate at the HNY-I and HNY,6X-G2 links. Unlike other heparinise, heparinase III does not contain the amino acid cysteine. It contains 13 histidine residues, which is responsible for its catalytic action.[2] This enzyme is not active towards heparin or low molecular weight heparins. Heparin-degrading lyase that recognizes heparin sulfate proteoglycan as its primary substrate. Source: Heparinase III from Flavobacterium heparinum Molecular Weight: ~70.8kD Specific Activity: 30units/mg protein (enzyme + BSA) Unit Definition: One unit will form 0.1umole of unsaturated uronic acid/hour at pH 7.5 at 25C, using bovine kidney heparan sulfate as substrate. Application: Heparinase III from Flavobacterium heparinum has been used to cleave specific GAGs.[3][4] It has been used to study its effect on the electrophoretic mobility of the protein reacting with the anti-aFGF (acidic fibroblast growth factor) antibody.[5] Storage and Stability: Lyophilized and reconstituted products are stable for 6 months after receipt at -20C. Reconstitute with sterile buffer or ddH2O. Aliquot to avoid repeated freezing and thawing. Store at -20C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
