Toxic component of a type II toxin-antitoxin (TA) system. Phosphorylates UDP-N-acetyl-D-glucosamine (UNAG) on the 3-hydroxyl group of the N-acetyl-D-glucosamine moiety, yielding UNAG-3P. UNAG-3P inhibits MurA, the first committed step in cell wall synthesis, which is then blocked. Phosphorylation is inhibited by cognate epsilon antitoxin. Part of a postsegregational killing (PSK) system involved in the killing of plasmid-free cells. The zeta toxin induces programmed cell death. Reconstitute with sterile ddH2O. Aliquot to avoid repeated freezing and thawing. Store at -20C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
Supplied as a lyophilized powder from 20mM Tris-HCl, 0.5M sodium chloride, pH 8.0, 6% trehalose. Reconstitute with sterile ddH2O to a concentration of 0.1-1mg/ml.
