Activin RIB, also known as ALK4, is a glycosylated 58kD type I transmembrane receptor that belongs to the superfamily of TGF-b serine/threonine kinase receptors. Activin RIB associates with Activin RIIB to form a receptor complex for activin and inhibin molecules (1). These ligands bind to Activin RIIB which then associates with, and phosphorylates, the cytoplasmic domain of Activin RIB to initiate signal transduction (2, 3). Mature mouse Activin RIB consists of a 103 aa extracellular domain (ECD), a 23 aa transmembrane segment, and a 356 aa cytoplasmic region that includes the kinase domain (4). Within the ECD, mouse Activin RIB shares 93% and 98% aa sequence identity with human and rat Activin RIB, respectively. It shares 23%-38% aa sequence identity with other mouse type I receptors Activin RIA, BMPR-IA, BMPR-IB, and TGF-b RI. Activin receptor signaling is modulated by the direct interaction of Activin RIB with cripto or inhibin binding protein (5-7). Activin RIB is excluded from the signaling complex if Activin RIIB first binds inhibin and betaglycan (8). Activin RIB functions in a wide variety of growth and differentiation processes, including embryonic cell fate and axis determination, cell proliferation, apoptosis, and tumorigenesis (1, 9, 10). A DNA sequence encoding the extracellular domain of mouse Activin RIB was fused to the Fc region of human IgG1 via a linker peptide. The chimeric protein was expressed in a mouse myeloma cell line, NS0. The recombinant mature mouse Activin RIB/Fc chimera is a disulfide-linked homodimeric protein. Based on N-terminal amino acid sequencing, the mature recombinant protein starts at Leu 32. The recombinant mouse Activin RIB/Fc monomer has a calculated molecular mass of 37.2kD. As a result of glycosylation, the recombinant monomer migrates as an approximately 44kD protein in SDS-PAGE under reducing conditions. Biological Activity: Measured by its ability to bind rmCripto. Immobilized rmActivin RIB/Fc at 1ug/ml (100uL/well) will bind rmCripto with a linear range of 0.8-100ng/mL. Endotoxin: 1.0 EU per 1ug of protein as determined by the LAL method. Storage and Stability: Storage and Stability: Lyophilized and reconstituted products are stable for 6 months after receipt at -20C. Reconstitute with sterile PBS. Aliquot to avoid repeated freezing and thawing. Store at -20C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
