| Membrane-bound form of TNF-a is cleaved by a convertase to generate a secreted 17kD mature protein. TNF-a converting enzyme, TACE, is a membrane bound metalloprotease-disintegrin. It also processes other cell surface proteins, including TNF receptor, TGFa, and alpha cleavage of amyloid protein precursor. Applications: Suitable for use in Immunohistochemistry, ELISA and Western Blot. Other applications not tested. Recommended Dilution: Peptide ELISA: 1:128,000 Western Blot: 0.1-0.3ug/ml, ~100kD band observed in Human Testis lysates and ~125kD in lysates of cell lines Daudi, HeLa and Jurkat (calculated MW of ~93kD according to NP_003174.3). Primary incubated for 1 hour. Preliminary testing was unsuccessful on Mouse and Rat Immunohistochemistry: 3.75ug/ml on paraffin embedded Human Skin and Thyroid Optimal dilutions to be determined by the researcher. Storage and Stability: May be stored at 4C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20C. Aliquots are stable for 12 months after receipt. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. |
