Human BRAF (a.a. 432-725) with N-terminal His tag, MW 36kD, co-expressed with p50 in an Baculovirus infected Sf9 cell expression system. Application: Suitable for use in the study of enzyme kinetics, screening inhibitors, and selectivity profiling. Other applications have not been tested. Recommended Dilutions: Optimal dilutions to be determined by the researcher. Specific Activity: As reported. Kinase Assay: Activity is tested using 1uM MEK1 (K97R) as substrate. Assay is performed in a 50ul reation in 10mM Tris, pH 7.4, 5mM magnesium chloride, 0.5mM EDTA, 0.5mM DTT, 5uM ATP, 5ug MEK1 (K97R) as substrate, and B2680-05A. 62ng of BRAF enzyme phosphorylated over 18% of 1uM MEK1 at RT in 1hr. Storage and Stability: Aliquot to avoid repeated freezing and thawing and store at -70C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Aliquots are stable for at least 6 months.
