Peptide is used to block beta-Catenin Rabbit mAb and beta-Catenin Antibody (Carboxy-terminal Antigen) reactivity in immunohistochemistry protocols. beta-catenin is a key downstream effector in the Wnt signaling pathway (1). It is implicated in two major biological processes of vertebrates: early embryonic development (2) and tumorigenesis (3). CK1 phosphorylates beta-catenin on Ser45. This phosphorylation event primes beta-catenin for subsequent phosphorylation by GSK-3 (4-6). GSK-3beta destabilizes beta-catenin by phosphorylating it at Ser33, Ser37 and Thr41 (7). Mutations in these phosphorylation sites, which result in the stabilization of beta-catenin protein levels, have been found in many tumor cell lines (8). Quality Control: The quality of the peptide was evaluated by reversed-phase HPLC and by mass spectrometry. The peptide blocks beta-Catenin Rabbit mAb and beta-Catenin Antibody (Carboxy-terminal antigen) by immunohistochemistry. Directions For Use: For immunohistochemistry, add twice the volume of peptide as volume of antibody used in 100ul total volume. Incubate for a minimum of 30 minutes prior to adding the entire volume to the slide. Recommended antibody dilutions can be found on the relevant product data sheet. Storage and Stability: May be stored at 4C for short-term only. For long-term storage, aliquot and store at -20C. Aliquots are stable for at least 6 months at -20C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
