Human cathepsin F (NM_003793), Cathepsin F (aa20-end), with C-terminal His-tag, MW = 53kD (more than 58kD after post-translational modifications), expressed in FreeStyle 293-F cells. Specific Activity: ~0.1pmole/min/ug. Assay Condition: Enzyme activity was measured in a buffer containing 100mM sodium acetate, pH 5.5, 2mM EDTA, 8mM DTT, 0.05% Brij, and 20uM substrate (Z-FRAMC), with 3.75-30ng/ul Cathepsin F. Reaction was done at 30C for 40-120 min, and fluorescence signal was measured at Ex360nm/Em460nm. Enzyme was preactivated in 50mM sodium acetate, pH 4.5, 5mM DTT buffer for 90 min at RT prior to reaction. Applications: Suitable for use in the study of enzyme kinetics, screening inhibitors and selectivity profiling. Other applications not tested. Recommended Dilution: Optimal dilutions to be determined by the researcher. Storage and Stability: Aliquot to avoid repeated freezing and thawing. Store at -70C. Aliquots are stable for at least 6 months at -70C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
